The Complexities of Linkage Studies for Neuropsychiatric Disorders
Abstract
Within the last decade there has been considerable fluctuation in opinion of whether genetic linkage was an appropriate paradigm for identifying major genes of psychiatric disorders and, even if appropriate, how such studies should be designed. The reviews presented by Baron (this volume) and by McGuffin et al. (this volume) are quite comprehensive and cover aspects of the history of the field as well as current perspectives on the major scientific issues and difficulties facing researchers in this field. While there are some who view the possibility of finding genes for psychiatric disorders by genetic linkage to be highly problematic at best, others, myself included, view it as the single most promising avenue of approach to these complex disorders. A promising line of research, however, is not necessarily simple and linkage studies of complex neuropsychiatric disorders are among the most difficult. Nevertheless, the effort is worthwhile given the individual suffering and extraordinary public health problems presented by these disorders. The variety of problems, difficulties, unresolved issues, and uncertainties raised by Baron (this volume) and McGuffin et al. (this volume) need not be restated. Rather, in the following paragraphs I will present a slightly different perspective on some of the relevant issues, provide some different examples relating to points they made, and, in at least one case, present a slightly different opinion.
