Scanning Transmission X-Ray Microscopy of Hydrated Mitotic Chromosomes

Abstract

Images of fully hydrated mitotic chromosomes from Vicia faba were recorded on the Scanning Transmission X-ray Microscope at 110, 85, 60, and 35 mM ionic strengths. The images revealed an increase in chromosome diameter and length as the salt concentration was reduced, in agreement with findings from visible light microscopy. Local differences in chromatin condensation were revealed in unstained chromosomes, which appeared as bands. Chromosome bands were most visible under salt conditions which support chromatin higher order structure, but not close packing of chromatin fibers.
