Comparison of Automated and Manual Methods of Liquid-Solid Sample Preparation for Determination of Drugs in Plasma by HPLC/UV
Abstract
Sample preparation schemes are often essential in the analysis of drugs in biological materials in order to concentrate the analyte within the detection limits of the instrument and remove potential endogenous interfering compounds. The most common method of sample preparation in the past has been liquid-liquid extraction, however liquid-solid extraction using chemically bonded silicas is becoming prominent1. Moreover, semi-automated methods such as the Advanced Automated Sample Processor (AASP) can give further gains in productivity in bio-analysis2. AASP methods comprise a manual off-line sample preparation stage followed by automated elution of the analytes by the mobile phase into a liquid chromatograph.
