The Human Immunoglobulin K Locus and its Evolution
Abstract
After an introduction on the evolution of the Summer Schools on Molecular Biology at Spetsai, the work of our group on the human immunoglobulin Kappa (K) locus is described. The locus consists of variable, joining and constant (VK, JK, CK) gene segments and comprises at least 2 Mb (mega-base pairs) of DNA. Up to now 81 of the perhaps about 100 VK genes and pseudogenes have been isolated on cosmid clones. Several contigs have been constructed covering about 1 Mb. An overall map of the locus is based on pulsed field gel electrophoresis experiments. The evolution of the K locus is described in terms of VK gene duplication, amplification, transposition and subgroup interdigitation. The role of pseudogenes within the K locus and of the pseudogenes which have been transposed to locations outside of the locus is discussed.
