Some Aspects of DNA Strand Separation

Abstract

Ever since the double helix structure was first proposed for DNA, the problem of strand separation during replication has attracted much attention. When I was asked by the organizers of this meeting to give a general survey on the physical chemistry of DNA, I decided to focus my discussions on strand separation. At the molecular level however, even the simplest in vitro DNA replication system is rather complicated, and little quantitative information is available. Therefore, the discussions to follow are qualitative and conceptual in nature.
