Propranolol-Induced Membrane Perturbation and the Metabolism of Phosphoinositides and Arachidonoyl Diacylglycerols in the Retina

Summary

The very active metabolic pathway for phosphatidic acid and phosphoinositides of the retina is affected greatly by the amphiphilic cation, propranolol. Propranolol, in addition to stimulating the synthesis of components of this pathway, also stimulates polyphosphoinositide degradation, probably due to the activation of a phospholipase C. Hence, propranolol stimulates the phosphoinositide cycle. This effect may be induced by a membrane perturbation caused by intercalation of the hydrophobic moiety of the drug in a membrane domain containing the components of the cycle. Exogenously added myo-inositol stimulates the synthesis of phosphatidylinositol and inhibits the propranolol-induced degradation of polyphosphoinositides suggesting that myo-inositol may modulate phosphoinositide metabolism. The use of propranolol as an experimental probe and the manipulation of phosphoinositide pathways by myo-inositol will prove useful in the study of the metabolism and function of phosphoinositides.
