Molecular cloning in heterologous systems

Abstract

During the last years the knowledge accumulated by fundamental research of DNA recombination in vitro became more relevant to biotechnology.

One of the main advantages of this so-called genetic engineering is the possibility to recombine genetic material from organisms which cannot be hybridized by means of the classical techniques of chromosomal recombination.

After a brief survey of the historical background of DNA transformation, the principal requirements of transforming DNA molecules are outlined, and widely used DNA vehicles are compiled.

Strategies and limits of molecular cloning are commented prior to the discussion of the main host organisms for foreign DNA.

In this review we have tried to select from the enormous number of results those which are of biotechnological relevance at present and those which may have future perspective in applied research.
