Abstract
Polyaromatic compounds which possess different base-pair selectivities in their DNA-binding (intercalating) properties were covalently bound to polyethylene glycol (PEG) via ester bonds. These PEG-bound intercalators are highly soluble in water and induced the retardation of DNA electrophoresis in agarose gel. The pattern of mobility retardation reflected the binding characteristics of the parent intercalators to DNA. Use of PEG-bound intercalators in the gel electrophoresis is discussed as a simple and convenient method for the characterization of polyaromatic DNA intercalators of low water solubility.

Binding equilibria were first studied with PEG(polymer)-acridine derivatives 5, 6 and 7. However, the rate at which the equilibrium is reached was so slow that the low-molecular compounds 8, 9 and 10 were employed as models.
The v values lower than 0.1 (less than one intercalator per every ten base-pairs) are considered to meet low loading conditions. The loading rate for compound 10 in Fig. 5 is appreciably higher than this criterion, but the plots afforded a straight line. The compound 9, though v≦1, gave concave plots, the first half of which (below v values of 0.05) was approximated to linear plots to obtain a rough measure of the binding constant.
