Arthrospira maxima (Spirulina) prevents endoplasmic reticulum stress in the kidney through its C-phycocyanin

极大节螺藻(螺旋藻) C-藻蓝蛋白阻碍肾脏内质网胁迫


极大节螺藻 (Arthrospira maxima) 细胞内含有C-藻蓝蛋白, 其具有抗氧化和细胞保护作用, 因此该藻全细胞和C-藻蓝蛋白均被确认为保健品. 然而, 它们的保健效应尚未被充分认识. 由于它们可以降低未折叠蛋白反应, 我们推测, 极大节螺藻细胞及其C-藻蓝蛋白可以防止肾脏内质网胁迫(ERS). 本文旨在研究极大节螺藻细胞及其C-藻蓝蛋白通过ERS反应对氯化汞 (HgCl2) 引起的肾损伤的影响. 实验公鼠先用极大节螺藻细胞或C-藻蓝蛋白处理30 min, 再用5 mg HgCl2/kg体重处理, 之后三天均饲喂藻细胞或C-藻蓝蛋白. 实验鼠安乐死后, 取其肾脏用于分析蛋白激酶RNA样内质网激酶(PERK), 转录激活因子 6(ATF-6) 和需肌醇酶 1α(IRE-1α) 信号途径. 结果表明, 汞中毒会提高PERK/eIF2α/ATF-4/GADD153和IRE-1α/XBP1 途径, 而极大节螺藻细胞处理防止了PERK, ATF-6α和半胱天冬酶的提高;同时, C-藻蓝蛋白处理使得所有这些蛋白得到正常表达. 本研究首次证实了极大节螺藻处理通过其C-藻蓝蛋白的活性防止了HgCl2引发的肾脏ERS.
