Development of a New RAM-Chiral Column for Determination of Chiral Drugs in Biological Fluids by HPLC

Abstract

New β-cyclodextrin (β-CD) bonded silica called RAM-Chiral has been designed for direct injection analysis of drug enantiomers in serum for liquid chromatography. The RAM-Chiral column is synthesized by two steps: introduction of a β-CD carbamate on a 70 Å pore size amino silica and coating of the external silica surface with BSA. The proteins were completely excluded and then recovered from the RAM-Chiral column. Direct injection analysis of benzodiazepin drugs in plasma can be determined on the RAM-Chiral column by using a step gradient buffered mobile phase. The recovery of racemic drugs from plasma was almost 100%.
