Abstract

A capillary zone electrophoresis (CZE) method with sensitivity cell (bubble cell) has been developed for the determination of sodium glucocorticoid hemisuccinates (Urbason® solubile forte 1000, Hydrocortison 100 Rotexmedica®, Prednisolut 100®) in biological media (plasma, urine). The samples in urine were diluted (1:1 v/v) in water and were injected into the apparatus without any further sample preparation. In plasma the formulations were also analyzed quantitatively. Here it was necessary to precipitate the protein component. Good results were achieved by treating with acetonitrile (1:3 v/v). The present results have shown that the investigation using a capillary with bubble cell led to an approximately 9-fold improvement of the detection limit compared to standard capillaries. The samples in the biological media were analyzed using a calibration curve for prednisolone hemisuccinate concentrations between 2.5 and 500 μg.mL−1. The detection limit and the relative standard deviation of the migration times and of the peak areas were determined.
