The effect of alendronate on bone tissue of senescent-accelerated OXYS rats

Abstact

The effect of alendronate on the bone tissue in male and female Wistar rats and senescent-accelerated OXYS rats with early development of osteoporosis was studied. The drug was applied at a dose of 0.84 mg/kg for 2 months starting from the age of 8 months. We found that the effect of the drug depended on genotype and sex of animals. It was maximally expressed in female OXYS rats as an increase in bond mineral density (BMD), which was initially decreased, and an elevation of the marker of bone formation osteocalcin in serum. In Wistar rats, the drug increased BMD in males only, whereas in females it decreased the osteocalcin level. The content of calcium in serum did not change and the known side effect of alendronate such as enhanced excretion of calcium with urine was expressed in male Wistar rats and in female of both strains. The response of OXYS rats to the application of the standard drug for osteoporosis treatment demonstrates that these rats can be used as a model of this disease, which is suitable for assessment of the efficacy of new therapeutic approaches.
