Mass spectrometry label-free quantitative analysis of proteins

Abstract

The dependence of the intensity of a mass spectrometric signal on protein concentration has been investigated using individual purified proteins and complex protein mixtures. Linearity of this dependence has been determined within the concentration ranges from 1 nM to 1 μM. In this range the dependence of the mass spectrometric signal intensity on concentration possesses a slope (tangent) characteristic for each protein studied. The efficacy of the method has been evaluated using the other methods (AQUA and 18O-labelling) employed in quantitative proteomics.
