Abstract
Neuromyelitis optica spectrum disorders (NMOSD) are autoimmune disorders affecting central nervous system with immunoglobulin G positivity to aquaporin-4 and myelin oligodendrocyte glycoprotein. NMOSD are clinically featured by transverse myelitis and optic neuritis. It is also a demyelinating autoimmune disease which could affect the erythrocyte deformability and may contribute to the severity of the disease. To study the electrophoretogram profile of erythrocyte proteins and its comparative expression between control and neuromyelitis optica variants. The study involves 4 groups: control, Aqp-4- IgG, MOG-IgG and NMO with autoantibody negativity to MOG and AQP-4 with samples of 3 per each group. The electrophoretogram of erythrocyte proteins was done using SDS PAGE and comparative band intensity between control and NMOSD variants was observed. The study observed there was increase in band intensity in the region between 37 KDa and 50 KDa in Aqp-4 IgG seropositive group compared to control, whereas no differential expression was observed in MOG-IgG positive variant. The erythrocyte proteins in those regions could be G3PD, actin and dematin. The bands in the region corresponding to 25 KDa were stomatin or Aqp-4 which showed marginal decrease in band intensity in NMO variants compared to control and the same observation was made in the region of 20 KDa where the band possibly represented the redox protein, peroxiredoxin. The differential band expression pattern of erythrocyte proteins between control and NMOSD variants could help in identification of red blood cell proteins which aid in the diagnostics and prognostics of NMOSD. According to comparative electrophoretogram the differentially expressed proteins could be G3PD, actin, stomatin and peroxiredoxin.
