The Biological Role of lncRNA linc01816 in Esophageal Squamous Cell Carcinoma and Its Correlation with Prognosis

Abstract

The present study aimed to explore the expression of linc01816 in esophageal squamous cell carcinoma (ESCC) and its effect on cellular activities of tumor cells, as well as its potential mechanism. RT-qPCR was used to detect the expression levels of linc01816 in human ESCC tissues and cell lines. ESCC cell lines were transfected with linc01816 siRNA and the difference in the proliferation abilities of different groups of cells was detected by CCK-8 method. Transwell method was used to detect the changes in its migration and invasion abilities. Kaplan–Meier curve was conducted to analyze the clinical value of linc01816 in ESCC. The relative expression of linc01816 was overexpressed in ESCC tissues and cells. High linc01816 expression was related to several clinical features and shorter overall survival rate. Compared with negative controls, linc01816 knockdown decreased the viability potential, migration and invasive abilities of ESCC cells, and was partially reversed by miR-326 inhibition. The overexpressed linc01816 was associated with ESCC patients’ overall survival. The proliferation and invasion capacities of ESCC cells can be inhibited by silencing linc01816, which may be related to the regulation of miR-326 expression. Linc01816/miR-326 axis may be a potential prognostic biomarker and therapeutic target for ESCC treatment.
