Zusammenfassung:
Der Nachweis von Barley yellow dwarf virus-Infektionen (BYDV) aus einzelnen Läusen mit Hilfe des ELISA-Tests war für den Einsatz im Routinetest keine Alternative, da die im Bestand gefundenen Läuse erst auf gesunde Pflanzen übersiedelt werden mussten, um dann eine Infektion an der Pflanze zu bonitieren. Dies war zu langsam und ungenau. Der im Pflanzenschutzamt Hannover angewandte One-tube-Immunocapture-RT-PCR-Test (RT-PCR) verbessert diesen Nachweis entscheidend. Es ist möglich, den Erreger aus Läusen, die bis zu vier Tagen in Auffanggefäßen einer Saugfalle gelegen haben, nachzuweisen. Bei Saugfallenfängen kann die einzelne Laus in Phosphatpuffer über mehrere Wochen eingefroren werden, um Untersuchungsengpässe zu überbrücken. Die Nachweissicherheit bleibt unverändert vorhanden. Ein Vergleich der Testmethoden ELISA und RT-PCR zeigte, dass mit Hilfe der RT-PCR wesentlich schneller eine Infektion in einer von Läusen beflogenen Pflanze nachgewiesen werden kann. Die sehr kurze Untersuchungsdauer von zwei Tagen bei diesem Testverfahren erlaubt es, kurzfristige und sicherere Prognosen über den weiteren Infektionsverlauf im Bestand zu stellen.
Summary:
The detection of a barley yellow dwarf virus infection in a single aphid by ELISA is not sufficient for a routine testing by the official plant protection service. First one have to transfer the aphids to healthy barley plants and after that the plants have been tested for the virus. This method needs a lot of time and is not reliable enough. For this reason the plant protection office of the chamber of agriculture in Hannover is using the one-tube- immunocapture-RT-PCR. With this testing method it is possible to detect BYDV-infections in single aphids which have laid up to four days in the liquid of vacuum traps. Aphids, which are caught by vacuum traps can be frozen in a phosphate buffer before testing. Even after several weeks the virus can easily be identified in the aphids. So an individual time management for carrying out the tests can be found by each laboratory. A comparison of the two test methods, ELISA and RT- PCR has shown, that the detection of a virus infection in plants can be done much more quicker by the RT-PCR than by ELISA. The very short testing time of about two days by RT-PCR allows to do a current and exact prognosis of the further infection development in the observed field.
