Abstract

CHARACTERISTIC of all known schemes of esterase catalysis1–5 is the assumption that the reactive form of serine pre-exists in the enzyme molecule before the interaction of enzyme with substrate or inhibitor. This seems to be unlikely, for why this very reactive form is not involved in side-reactions is still very vague. Moreover, the possibility of the preliminary activation of serine aliphatic hydroxyl, by means of such a weak base as imidazole3–5, is not convincing.
