Oxidative Phosphorylation in the Chick Embryo

Abstract

SYNTHESIS of high-energy phosphate bonds was measured by the estimation of phosphorus: oxygen ratios. The materials used were 7–9 day old chick embryos of a hybrid White Leghorn ♀ × New Hampshire Red ♂. Except for the step isocitrate→ succinate, the estimations were done on the mitochondrial fraction, prepared according to the method of Hogeboom and Schneider1. Oxygen consumption was measured by the Warburg technique, using 0.1 M phosphate buffer at pH 7.4 with the addition of adequate co-factors and substrates2. Phosphorus estimations were carried out according to the method of Lowry and Lopez3.
