Abstract
Pealius mori Takahasshi is an important pest of mulberry trees, but little is known about its distribution and genetic differentiation. Eighteen populations of P. mori were collected from different sericultural areas in Yunnan Province. They were analyzed using mitochondrial cytochrome c oxidase I (mtCOI) subunit sequence cluster analysis. There were seven clades of P. mori present in Yunnan. Most P. mori in southeast, central, northeast, southwest, and west Yunnan were clade I, which was the dominant species of pests. Another potential dominant species was clade II, which was from east, northwest, and central Yunnan. P. mori of clades III–VII were sporadically distributed in various sericulture areas. The genetic diversity of P. mori was high, as indicated by the total haplotype diversity (Hd = 0.942), nucleotide diversity (Pi = 0.11734), and average nucleotide difference (K = 77.207). The higher genetic differentiation index (FST > 0.70) and smaller gene flow (Nm < 1) in most populations represented a higher differentiation degree. Tajimma's D and Fu and Li's D* neutral tests indicated that 14 populations of P. mori had population expansion, and 10 of these were limited to local areas. Analysis of the molecular variance of P. mori showed that the genetic variation mainly came from different regions (54.97%), and the variation within populations was the smallest (16.40%). Mantel test showed no correlation between genetic and geographic distance (r = 0.0667, P = 0.2490).
