Summary
Fig tree is commonly affected by fig mosaic disease (FMD), the symptoms of which include vein clearing, necrotic edges, mosaic and leaf deformation. FMD has been associated with at least twelve viruses and three viroids. Fig plant leaves with FMD symptoms were collected in Mexico in two nurseries located in Texcoco and Tetela del Volcán, respectively. PCR tests, using specific primers for phytoplasmas and PCR/RT-PCR analysis for nine viruses and three viroids, were performed. Three viruses were detected, including fig mosaic emaravirus (FMV, 64%), fig badnavirus-1 (FBV-1, 53%), and fig leaf mottle-associated virus 1 (FLMaV-1, 7%). Mixed infections were also detected, including FMV + FBV-1 (32%) and FMV + FLMaV-1 (7%). The results of phytoplasma sequencing, phylogenetic and RFLP analysis indicated the presence of ‘Candidatus Phytoplasma asteris’ (46.4%) belonging to the 16SrI-B subgroup. In leaves showing chlorotic mottling, no viruses or phytoplasmas were detected. Viroids were not found in any of the samples.
