Abstract
The polyherbal preparation (PHP) was screened for its antiosteoporotic potential in ovariectomized rats. To design herbal amalgam, extraction of Withania somnifera, Cissus quadrangularis, and Pinus gerardiana was done using Soxhlet extraction. The PHP was standardized using HPLC profiling. The antiosteoporotic activity was accessed using ovariectomized female Wistar rats 180–250 g. PHP was given at two doses: 100 and 200 mg/kg and Reosto at 250 mg/kg (standard marketed formulation) orally for 35 days. Further, phytocompounds from selected plants were docked with osteoporotic protein targets Cathepsin, Receptor activator of nuclear factor κ B ligand (RANKL), Osteoprotegerin (OPG). The findings from the HPLC study revealed the presence of eighteen compounds among which gallic acid, quercetin, and rutin were verified. There was a significant elevation in serum calcium, phosphorus, estradiol levels while serum ALP level was found to be decreased resulting in the restoration of bone mass. Somniferine, withaferin A, and β-sitosterol showed a strong affinity with all the target proteins thereby indicating their potential against osteoporosis. Taken together, our findings indicated that PHP has the efficacy to ameliorate the pathological state of osteoporosis and may be used as an alternative drug for osteoporosis.

Significance Statement In the present study antiosteoporotic effect of a polyherbal preparation in ovariectomized rats was studied. PHF was prepared by extraction of W. somnifera roots, C. quadrangularis stem, and P. gerardiana bark. The PHF was standardized using HPLC profiling. Phytocompounds were docked with osteoporotic protein targets Cathepsin, RANKL, and OPG.
