Abstract
Brucellosis is a major bacterial zoonosis affecting domestic livestock, wild mammals, as well as humans around the globe. In the present study, proteomes of three different species of Brucella were compared by two dimensional electrophoresis (2-DE). B. abortus, B. melitensis and B. suis, were cultured in Mueller–Hinton agar. The cells were harvested by centrifugation and lysed in lysis buffer. Protein fraction was purified by 2-DE cleanup kit and subjected to isoelectric focusing (IEF) using 3–10 pH gradient Immobiline dry strips. After IEF, the strips were equilibrated and subjected to 2nd dimension SDS PAGE. The gels were stained with Coomassie Brilliant blue stain, scanned and analysed using Image master platinum 7. A total of 381, 386 and 362 protein spots were detected for B. abortus, B. melitensis and B. suis, respectively. Out of them, 312 spots (proteins) were common to all the three species of the bacteria, the remaining spots were unique either to one species or two, but were not present in all the three species. Twenty-two proteins were significantly over-expressed in B. abortus as compared to B. melitensis and B. suis, while 12 proteins were significantly over-expressed in B. melitensis as compared to the rest of the species.
