Isolation and characterization of 24 microsatellite loci in oyster Crassostrea hongkongensis

Abstract

Twenty-four polymorphic microsatellite markers were developed from the Hong Kong oyster, Crassostrea hongkongensis, with a partial genomic library enriched for tandem repeat sequences of (CA)12, (GA)12. Polymorphism of these loci was evaluated in a sample of 34 wild unrelated individuals. At these polymorphic loci, the number of alleles per locus vary from 4 to 12, with the average of 7.17; Observed heterozygosity vary from 0.412 to 0.912 with an average of 0.674, while expected values for these loci range from 0.698 to 0.899 with an average of 0.822. After Bonferroni correction (P < 0.002), departure from Hardy–Weinberg equilibrium are observed in four loci (Ch210, Ch211, Ch214 and Ch217) due to homozygote excess and not detected in others. Additionally, four loci (Ch209 and Ch211, Ch207 and Ch224) showed significant linkage disequilibrium (P < 0.01). This set of microsatellite loci would be useful for stock and genetic studies in C. hongkongensis.
