Abstract
Protein-protein interactions are key elements of intracellular signalling and metabolic pathways. These interactions can be revealed with the help of proximity ligation screens, prominently biotinylation screens. This approach has profited from the recent development of the highly active biotin ligase TurboID, which however also led to problems of toxicity related to its high basal activity. We have established a simple protocol to improve TurboID performance and enhance protein functionality.
