Fertility conversion between cytoplasmic maintainer lines and restorer lines through molecular marker-assisted selection in pepper (Capsicum annuum L.)

Abstract

Pepper commercial hybrids, produced by an economical and efficient cytoplasmic male sterility (CMS)/restorer-of-fertility (Rf) system, have been widely used in production because of their strong heterosis. However, the restorer resources of pepper are rare in nature, which makes it necessary to transfer the Rf gene from restorer lines to maintainer lines to produce hybrid F1 seeds. Here, we backcrossed a core maintainer line ‘247’ (receptor) with a three-line hybrid pepper ‘17–120’ (donor) to transfer the Rf gene into the maintainer line. Two molecular markers, CRF and SCD06-17, linked to the Rf gene were used to screen individual plants with the Rf gene (foreground selection) and 31 SSR markers were used in BC1F1 and BC2F1 generations to screen individuals with the highest similarity to the genetic background of the recurrent parent (background selection). Restorer line ‘247R’ was acquired in four generations of breeding and did not differ significantly with the recurrent parent between the main agronomic traits. The scheme used in this study accelerated the fertility conversion process between maintainer lines and restorer lines, which lays a foundation for the production of hybrid seeds by maintainer lines.
