A repertoire of anti-mannan Candida albicans antibodies in the blood sera of healthy donors

Abstract

Fungal mannans are polysaccharide antigens located on the cell wall surface of various representatives of the fungal kingdom. Titers of the IgG antibodies to Candida albicans mannan are determined in the diagnosis of the candidiasis infection. Owing to high complexity and heterogeneity of the structure of natural mannans, the determination of the exact epitope structure recognized by antibodies is extremely important for the improvement of diagnostic tools based on the detection of anti-mannan antibodies. A high level of the antibodies to mannan was also found in healthy donors. However, it remains undetermined what structural epitope was recognized in this case. The levels of the IgG and IgM antibodies against individual structural fragments of mannan in the blood sera of healthy donors were determined. The anti-mannan antibody response was measured using enzyme immunoassay in a random sample of 31 blood sera of healthy donors on a panel of 11 synthetic mannooligosaccharides of various structures. A high antibody titer was found for the antigens containing the β-mannosidic bond, and the highest values were achieved in the case of two (1→2)-linked β-mannose residues. No significant level of the antibodies was revealed for antigens containing only α-mannose residues. Thus, it can be concluded that β-mannans appear to be much stronger antigens. It is assumed that the detection of antibodies to β-mannans in the sera of healthy donors can be a source of false positive results in the serological diagnosis of the Candida infection.

D. Zelinsky Institute of Organic Chemistry of RAS (Moscow). A candidate to Academicians of RAS (elections of 2022). The main studies of N. E. Nifantiev are devoted to the chemistry and conformational analysis of oligo- and polysaccharides, investigation of glycosylation mechanisms, and development of methods for stereo- and regiodirectional building of the glycosidic bond and carbohydrate bond assembling, including vicinal branched and polyanionic compounds that are most difficult for investigation. These studies made it possible to prepare original vaccines, infection diagnosticums, and drugs. Among them are (i) vaccines against Staphylococcus aureus (now under international tests), Haemophilus influenzae (required for the National Immunization Schedule for Children Protection; the production was organized in cooperation with the ROSTEKH State Company), and a number of other bacterial and fungal pathogens; (ii) diagnosticums of infections, including the single domestic diagnosticum of invasive aspergillosis that is anticipatory of the best foreign analogs in specificity (registered in 2019, used in the RF, EU, and Asia); (iii) hemapoiesis stimulator, cardioprotector, and antiviral and other drugs being at different stages of testing. The drug form of the photosensitizer providing its accumulation in a tumor was developed: the drug is allowed for the treatment of head and neck squamous cell carcinoma in several countries. N. E. Nifantiev is the author and coauthor of more than 400 articles, 25 reviews, and 50 patents. N. E. Nifantiev is a member of the Bureau of the Division of Chemistry and Materials Science of RAS and Editorial Boards of Vestnik RAN (RAS Bulletin), Bioorganic Chemistry (Bioorganic Chemistry of RAS), Russ. Chem. Bull. (RAS), Journal of Carbohydrate Chemistry, and four other domestic and international journals; Vice-President of the Scientific Council of RAS on Organic Chemistry; President (2020–2021) and Past President (2022–2023) of the Division of Organic and Biomolecular Chemistry of IUPAC; Chairman of the subcommittee “Organic Synthesis” of IUPAC; and a representative of RAS at the International Council of Science (ICS, earlier ICSU). N. E. Nifantiev was awarded with the Leninsky Komsomol Prize (1989, for the works on stereodirectional synthesis of polysaccharides) and the RAS Prize named after M. M. Shemyakin (2013, for the cycle of works “Stereodirectional Synthesis of Carbohydrate Chains of Glycolipids and Glycoproteins and Their Conjugates for Glycobiological Research”).
