Abstract
The nearly isogenic lines of Cucurbita pepo L designated 7-11-1 and 7-11-2, which possess characters of smooth-edged seeds and reductus-edged seeds, respectively, were subjected to complementary DNA (cDNA)-randomly amplified polymorphic DNA (RAPD) technology to assess differential gene transcription in seed coats. Gene fragments showing differential expression were obtained. Full-length mRNA was recovered by rapid amplification of cDNA ends and the corresponding gene designated GSE-1. BLAST analysis in internet database indicated that this is a novel gene of 1,225 bp contains an open reading frame (ORF) of 903 bp and two typical zinc finger domains. Amplification of this gene from two different inbred lines indicated that the 55th nucleotide of the ORF in the 7-11-2 inbred line had mutated from C to T, thereby terminating the ORF prematurely. Spatial and temporal analysis of gene expression by RT-PCR indicates that GSE-1 is a novel gene specifically expressed in seed coats that displays temporal specificity during development.
