Production and Properties of Aspergillus fumigatus Collagenolytic Proteinase

Collagenolytic proteases (collagenases) can be used in a variety of areas of human life: processing of leather, production of detergents, and food and beverage preparation, as well as pharmaceuticals and medicine. The aim of this study was to develop methods for the preparation and characterization of collagenolytic proteinase from the filamentous fungus Aspergillus fumigatus. A method for preparing A. fumigatus extracellular collagenolytic proteinase by two-stage chromatographic purification was developed. Analysis of the physicochemical and biochemical properties of the enzyme confirmed that a serine-type proteinase with a molecular weight of 40 kDa and maximum activity at 40°C, pH 7.5, was synthesized. The data obtained here suggest that A. fumigatus proteinase can be regarded as a basis for obtaining new drugs with collagenolytic effects.
