Diuretic and Neurotropic Activity of Oreganol A, a Component of Oregano

Oregano (Origanum vulgare L.) is a valuable source of biologically active substances, including phenolic compounds. The aim of this study was the isolation of 3′,4′-dihydroxybenzylprotocatechoic acid 4′-O-β-D-glucopyranoside from O. vulgare herb and subsequent analysis of its nephrotropic and neurotropic activity. Pure 3′,4′-dihydroxybenzylprotocatechoic acid 4′-O-β-D-glucopyranoside (oreganol A) was isolated using column chromatography over L 40/100 silica gel and identified using UV, PMR, and 13C NMR spectroscopy and mass spectrometry. Diuresis significantly increased in animals of the experimental group relative to the water control indicators with a single intragastric administration of oreganol A at a dose of 0.5 mg/kg against a background of 3% water load in a 4-hour experiment. Diuresis and creatininuresis significantly increased in animals of the experimental group by the end of a 24-hour experiment. An analysis of the results of a Porsolt swimming test revealed that the motor activity of experimental animals relative to a water control decreased significantly with a single intragastric injection of oreganol A at a dose of 0.5 mg/kg.
