Synthesis and Anti-Inflammatory Activity of N,6-Diaryl-4-Methyl-2-Thioxo-1,2,3,6-Tetrahydropyrimidine-5-Carboxamides

Previously unreported N,6-Diaryl-4-methyl-2-thioxo-1,2,3,6-tetrahydropyrimidine-5-carboxamides were synthesized by reacting acetoacetanilides with a mixture of an aromatic aldehyde and thiourea. The anti-inflammatory activity of the series of synthesized compounds was assessed. The anti-inflammatory activities of 4-methyl-N,6-diphenyl-2-thioxo-1,2,3,6-tetrahydropyrimidine-5-carboxamide and 4-methyl-N-phenyl-6-(2-chlorophenyl)-2-thioxo-1,2,3,6-tetrahydropyrimidine-5-carboxamide, which inhibited the inflammation reaction in a rat-paw edema model by 79.54 and 79.84%, respectively, were statistically significantly greater than that of nimesulide (55.74%).
