PMAA nanogel controllably releases anti-IL-1β IgY for treating allergic rhinitis

Abstract

Intranasal glucocorticoid spray is the preferred method to treat allergic rhinitis. However, due to the instantaneous high drug concentration, there are still some side effects, such as nasal dryness, nasal bleeding and even systemic effect of glucocorticoid. Herein, a novel biocompatible poly(methacrylic acid) (PMAA) nanogel was synthesized and used to load anti-IL-1β IgY (IgY) for treating allergic rhinitis induced by ovalbumin. The results indicated that PMAA nanogel had sustained-released effect of IgY in vitro. Symptoms of allergic rhinitis were remarkably relieved after the topical intranasal drip treatment using IgY/PMAA nanogel. The number of eosinophilic granulocyte in nasal lavage fluid and in nasal mucosa tissue, the expression of IL-1β analysis revealed that IgY/PMAA nanogel had a good controlled-released therapeutic effect, and IgY/PMAA nanogel exhibited lower in vivo toxicity in the experimental range, which suggested a promising potential for allergic rhinitis treatment with less side effect.
