Fluorescence of Hoechst 33258 Complexes with Serum Albumin and DNA

Fluorescence spectroscopy was used to study the interactions of the bisbenzimidazole compound Hoechst 33258 with bovine serum albumin and calf thymus DNA. The fluorescence spectra of these complexes are presented. The maxima in these spectra were used to construct curves for the dependence of the fluorescence intensities of this ligand bound to the indicated protein and DNA on the macromolecule/ligand ratio r. In the case of albumin, the intensity decreases with increasing protein concentration in the low protein concentration region and then increases with increasing protein concentration in the region of higher protein concentrations. In the case of DNA, the intensity steadily increases with increasing DNA concentration. This behavior is attributed to binding of the ligand to both DNA and albumin.
