Alcohol treatment promotes conversion of testosterone to estradiol-17β in female tilapia, Oreochromis mossambicus

Abstract

Effect of ethanol treatment on production of sex steroid hormoneswas assessed in the present study. Intraperitoneal administration (1.16 μl/g body weight) of ethanol to mature female tilapia, Oreochromis mossambicus, had little effect on plasma level of estradiol-17β (E2), but resulted in a significant decrease in plasma level of testosterone (T) at 6 days after administration. Ratio of E2 to T increased significantly from 3 days after ethanol administration. When pieces of ovaries from mature female tilapia were incubated with different doses of ethanol (0, 0.58, 2.9 and 5.8 μl/ml medium), significantly higher E2 and lower T productions were observed at 48 h in the higher ethanol treatments (2.9 and 5.8 μl/ml medium). This steroid production at higher ethanol concentration coincided with a significantly higher cytochrome P450 aromatase gene (P450arom) mRNA abundance as assessed by reverse transcription-polymerase chain reaction. Taken together, these results suggest that alcohol activates P450arom activity and promotes the conversion of T to E2 in the ovary of tilapia.
