Isolation of Sesquiterpene Lactones and the Antioxidant and Anticancer Activities of Crude Extracts from Cyanthillium cinereum

Three sesquiterpene lactones has been isolated from Cyanthillium cinereum (L.) H. Rob. by bioactivity guided assay and characterized by spectral methods as compounds 1, 2, and 3. The in vitro anticancer activity was tested against 786-O, MCF-7, and K562 cells by the MTT assay. Compound 1 showed IC50 values of 12.5 and 10.4 μg/mL against 786-O cell line after 48 and 72 h, respectively, and matches with doxorubicin′s IC50 values of 12.2 and 9.1 μg/mL, respectively, in the same time period. The antioxidant activity by the DPPH assay showed IC50 values of 45.0, 48.0, and 44.7 μg/mL for the three compounds. Both cell cycle analysis and LDH release assays revealed that compound 1 affects DNA replication by inducing a block at the S phase. Compound 1 treatment led to increased levels of ROS production and could be an intermediate step in inducing cytotoxicity.
