Fig. 3 The interaction between nNOS and OGT was recently reported (Ryu et al. 2016). As OGT catalyzes the OGlcNAc modification of nNOS during excitotoxicity, we speculated whether nNOS interacts with OGT. Immunofluorescent staining showed that nNOS was strongly co-localized with OGT 3 h after ischemia compared with the sham group (ag). In addition, nNOS directly interacts with OGT in brain tissue as indicated by reciprocal immunoprecipitation studies (h, i). Subsequently, immunoprecipitation studies showed that in PC12 cells treated with or without glutamate, nNOS interacted with OGT directly, and the interaction between nNOS and OGT was enhanced by glutamate stimulation as well as O-GlcNAcylation of nNOS (j). Thus, we have sufficient reason to conclude that excitotoxicity induces the increase in nNOS O-GlcNAcylation due to an enhanced interaction between nNOS and OGT
