Rufinamid und Stiripentol – zwei neue Antiepileptika und die Premiere von Orphan drugs in der Epilepsietherapie

Abstract

Rufinamide and stiripentol are the two most recently licensed anticonvulsants in Germany. The peculiarity consists of the approval as so-called orphan drugs. For an adequate risk-benefit consideration it is helpful to know the special features of such an approval. Rufinamide is approved for the treatment of Lennox- Gastaut syndrome, stiripentol for Dravet syndrome. The specific advantages and disadvantages of orphan drugs in epilepsy treatment are discussed.

Zusammenfassung

Rufinamid und Stiripentol sind die beiden zuletzt in Deutschland zur Epilepsietherapie zugelassenen Antikonvulsiva. Die Besonderheit besteht darin, dass beide den besonderen Zulassungsstatus als so genannte Orphan drugs haben, also als Arzneimittel für seltene Krankheiten, aufweisen, dessen Eigenheiten zu kennen für die therapeutische Nutzen-Risiko-Abschätzung bedeutsam ist. Rufinamid ist für die Therapie des Lennox-Gastaut-Syndroms, Stiripentol für die Behandlung von Patienten mit Dravet-Syndrom zugelassen. Die spezifischen Vor- und Nachteile von Orphan drugs in der Epilepsiebehandlung werden diskutiert.
