Résumé

Les substances naturelles issues des végétaux présentent des intérêts multiples mis à profit dans la biotechnologie tant dans l’industrie alimentaire, cosmétique que pharmaceutique. Parmi ces composés, on retrouve une grande partie des métabolites secondaires qui se sont illustrés dans beaucoup de domaines et même en thérapie. Dans ce contexte, nous avons recherché l’activité antioxydante de Berberis vulgaris, une plante utilisée en Algérie et plus particulièrement dans la région de Tlemcen. Après préparation des extraits à partir des écorces de racines de Berberis vulgaris, les alcaloïdes totaux ont été analysés par chromatographie sur couche mince, révélant la présence de six taches. Aussi, une analyse quantitative des polyphénols et des flavonoïdes de l’extrait hydrométhanolique a été réalisée, montrant une teneur de 10,48 mg GAE/g en polyphénols totaux et 2,05 mg CEQ/g en flavonoïdes. L’activité antioxydante des extraits hydrométhanolique et alcaloïdes totaux a été évaluée par deux méthodes: la réduction du fer et le piégeage du radical libre DPPH. Les résultats obtenus ont montré une activité élevée de l’extrait hydrométhanolique par rapport aux alcaloïdes totaux, avec une CI50 de 1,40 mg/ml.

Abstract

Natural substances from plants have multiple interests exploited in biotechnology industry both in food, cosmetic and pharmaceutical. These compounds include a large proportion of secondary metabolites that are illustrated in many areas and even in therapy. In this context, we investigated the antioxidant activity of Berberis vulgaris, a plant used in Algeria, and it is particularly used in the Tlemcen region. After preparation of extracts from the root bark of Berberis vulgaris, total alkaloids were analyzed by thin layer chromatography, indicating the presence of six tasks. A quantitative analysis of polyphenols and flavonoids in methanolic extract was carried out, showing a content of 10.48 mg GAE/g of total polyphenols and 2.05 mg CEQ/g of flavonoids. The antioxidant activity of hydromethanolic extracts under reflux and total alkaloids was evaluated using two methods: reducing power and DPPH free radical scavenging. The results showed a high activity of methanolic extract than the total alkaloids, with an IC50 of 1.40 mg/ml.
