Representative DEAE-Sepharose Fast Flow elution profile of radish leaf kaempferitrin α-rhamnosidase activity. Leaves sampled from radishes stored at 5 °C for 5 days were homogenized with enzyme extraction buffer, protein precipitated with ammonium sulfate and desalted on a Sephadex G25 column as described under Materials and methods. Thereafter, the α-rhamnosidase activity was loaded onto a DEAE-Sepharose Fast Flow column, and protein eluted with increasing [NaCl].
