PF presynaptic LTD required the activation of CB1 receptors and is induced by the application of a CB1 agonist. a LTD of the PF Ca2+ transients was prevented by the CB1 receptor antagonist AM251 (5 μM). b Bath application of the CB1 agonist WIN55212-2 (2 μM, 10 min) induced a depression of PF Ca2+ transient that outlasted agonist application (filled circles,WIN55212-2, n = 6) and persisted when the CB1 antagonist AM251 was applied after WIN55212-2 (open circles, WIN55212-2→AM251, n = 6). WIN55212-2 was ineffective when the CB1 antagonist AM251 was applied before WIN55212-2 (open squares, AM251→WIN55212-2, n = 6). c LTD of the PF Ca2+ transients was prevented by the neutral CB1 receptor antagonist O-2050 (10 μM). d WIN-induced depression of PF Ca2+ transients was not augmented by subsequent 4 Hz PF stimulation (n = 6).
