Zusammenfassung
Als Risiken für die Entwicklung einer rheumatoiden Arthritis (RA) sind sowohl umweltbedingte als auch genetische Faktoren anzusehen. Die RA, als chronische Entzündung, gilt darüber hinaus selbst als Risikofaktor für die koronare Herzerkrankung (KHK), da sie – oder ihre Behandlung – eine endotheliale Dysfunktion verursachen kann. Ein Polymorphismus im Promoter der endothelialen NO-Synthase (–786C/T) führt bei –786CC-homozygoten Personen ebenfalls zu endothelialer Dysfunktion. Dieser Gendefekt ist vermehrt bei Patienten mit RA anzutreffen und daher als genetischer Risikofaktor für RA anzusehen.
Abstract
The risk to develop rheumatoid arthritis (RA) includes environmental as well as genetic factors. Moreover, RA, being an inflammatory disease, is itself considered to be a risk factor for coronary artery disease, as it – or its treatment – may lead to endothelial dysfunction. Likewise, a single nucleotide polymorphism in the promoter of the endothelial NO synthase (–786C/T) leads to endothelial dysfunction in –786CC homozygous individuals. This genetic defect occurs in significantly higher frequencies in patients with RA, and therefore must be regarded as a genetic risk factor for RA.
