S-Antigen (AgS) und Interphotorezeptor-retinoid-bindendes Protein (IRBP) sind 2 hochpotente uveitopathogene Autoantigene der Retina, die im Tierexperiment häufig zum Triggern einer Autoimmunuveitis (EAU) eingesetzt werden.
Ziel der Untersuchungen war die simultane Isolierung beider Proteine aus boviner Retina unter Vermeidung zeitaufwendiger Dialyse und Verwendung kleiner, kommerziell verfügbarer Fertigsäulen.
Methode: Retinaextrakt wurde nach bekanntem Modus isoliert und mit Ammoniumsulfat gefällt. Entsalzung und Umpufferung des gelösten Präzipitats wurden an einer Bio-Gel P6-Säule durchgeführt. Die anschließende Fraktionierung an Econo-Pac Q lieferte eine AgS- und IRBP-Rohfraktion, deren weitere Reinigung vorwiegend an 5-ml-Econo-Pac Q, -HTP- und -HIC-Kartuschen erfolgte.
Ergebnisse: AgS und IRBP wurden in hoher Reinheit (Natriumdodecylsulfat-Polyacrylamidgelelektrophorese, Silberfärbung) aus boviner Retina unter Verwendung kleiner Fertigsäulen und mit Ausbeuten von etwa 0,25 (AgS) bzw. 0,15 (IRBP) mg/g Retina isoliert.
Schlußfolgerung: Fertigkartuschen ermöglichen die simultane Isolierung von AgS und IRBP in mg-Mengen unter methodisch vereinfachten Bedingungen. Dieses Vorgehen könnte insbesondere bei begrenzter Retinamenge interessant sein.
S-antigen (AgS) and interphotoreceptor retinoid-binding protein (IRBP) are two highly potent uveitopathogenic autoantigens of the retina frequently used to trigger autoimmune uveitis in animal trials.
Purpose: The aim of this study was the simultaneous isolation of both proteins from bovine retina, additionally avoiding time-consuming dialysis and employing prepacked, commercially available cartridges.
Method: Retina extract, obtained in the usual manner, was precipitated with ammonium sulfate. Both the desalting and the buffer exchange of dissolved precipitate were carried out using a Bio-Gel P6 column. Subsequent separation with Econo-Pac Q gave prepurified AgS and IRBP fractions. Further purification was realized predominantly with Econo-Pac Q, -HTP and -HIC (5-ml cartridges).
Results: AgS and IRBP were isolated in high purity (sodium dodecylsulfate polyacrylamide gel electrophoresis, silver stain) from bovine retina using prepacked cartridges with yields of approximately 0.25 (for AgS) and 0.15 (for IRBP) mg/g retina, respectively.
Discussion: The application of ready-to-use cartridges allows simultaneous isolation of AgS and IRBP in milligram amounts under simplified conditions. This approach might be of particular interest for small samples of retina.
