Transdifferenzierende retinale Pigmentepithelzellen (RPE) sind zunehmend kontraktionsfähig und gelten als kritischer Faktor in der Genese traktionsbedingter Netzhautablösungen. Ziel dieser Studie war es, Faktoren nachzuweisen, die einen Kontraktionsreiz propagieren, und einige beteiligte Mechanismen zu beschreiben sowie Inhibitoren zu finden. Unter Verwendung eines In-vitro-Kontraktionsmodells konnten Serum, Platelet-derived-growth-Faktor (PDGF) und Insulin-like-growth-Faktor-1 (IGF-1) als starke Kontraktionsinduktoren ermittelt werden. Endothelin 1 (ET-1), Transforming-growth-Faktor (TGF)-β1 und -β2 sind weniger effektiv. Der durch Serum vermittelte Kontraktionsreiz wird sowohl direkt als auch indirekt (Proteinsynthese-abhängig) propagiert. Die durch Peptide induzierte Kontraktion erfolgt indirekt über die Syn-these neuer Proteine und kann durch Cycloheximid vollständig blockiert werden. Proteinkinasen nehmen innerhalb der Kontraktionsmechanismen eine zentrale Stellung ein und können durch Staurosporin, ein Proteinkinasehemmer, effektiv inhibiert werden.
Transdifferentiated retinal pigment epithelial cells (RPE) display enhanced contractile potentials and have been implicated in the development of tractional retinal detachment. This study determines the activity of contraction-promoting factors, examines some involved mechanisms and evaluates inhibitors. Using an in vitro contractionassay, we demonstrated that collagen matrix contraction by transdifferentiated RPE cells is effectively stimulated by serum, platelet-derived growth factor and insulin-like growth factor-1. Endothelin-1 and transforming growth factor-β1 and -β2 have a more discrete or marginal effect. Tractional forces promoted by these peptides are completely protein synthesis dependent. Contraction stimulated by serum is only partly dependent on de novo protein synthesis, suggesting different active factors and/or pathways. Staurosporine, a broad-spectrum kinase inhibitor, effectively inhibited collagen matrix contraction by transdifferentiated RPE cells regardless of the promoter.
