Angiotensin-I-converting-Enzym (ACE, CD143) in regelrechtem humanem Lungengewebe

Zusammenfassung

Angiotensin-I-converting-Enzym (ACE, CD143) ist eine von Endothelzellen transmembran exprimierte Zn2+-Dipeptidylpeptidase, die durch Bildung von Angiotensin II und Degradation von Bradykinin vasokonstriktorisch wirkt. Wir überprüften immunhistochemisch die endotheliale ACE-Expression in der humanen pulmonalen Strombahn in Lungengewebeproben von 39 Patienten (20 Frauen, 19 Männer; 34–76 Jahre). In allen Proben zeigten die pulmonalkapillären Endothelzellen die stärkste Expression, gefolgt von denen der Arteriolen und Arterien. In Venolen sowie Venen fand sich keine nennenswerte ACE-Expression. Statistisch signifikant waren die Intensitätsunterschiede zwischen den Gefäßtypen (p<0,001), jedoch nicht bezüglich des Geschlechts. Es kann somit ein gefäßtypspezifisches ACE-Expressionsmuster der pulmonalen Strombahn belegt werden. Dies weist ACE als endothelialen Marker für diese Lungengefäße aus.

Abstract

Angiotensin I converting enzyme (ACE, CD143) is an endothelial transmembrane Zn2+-dipeptidylpeptidase. By formation of angiotensin II and degrading bradykinin it acts as a vasoconstrictor. We examined endothelial ACE expression in human pulmonary vessels in specimens from 20 female and 19 male patients (age: 34–76 years) by immunohistochemistry. In all specimens, capillary endothelial cells showed the strongest expression, followed by those in arterioles and arteries. Venules and veins showed next to no staining. The differences in staining intensities were significant (P<0.001). Sex affected neither the expression intensity nor the expression pattern. Summarizing, we demonstrate the existence of a vessel-type specific ACE expression pattern for pulmonary vessels. The nearly exclusive endothelial ACE expression in capillaries and arterial vessels points to ACE as an immunohistochemical marker for these vessels in normal lung tissue.
