A number of publications have reported the in vitrotoxicities of local anesthetics and steroids commonly injected into joints on cultured chondrocytes. In this study, fibroblast, tenocyte and human mesenchymal stem cell (hMSC) cultures were incubated for 30 min while exposed to ropivacaine, bupivacaine or triamcinolone at a 0.125%, 0.25% or 0.5% concentration. The agents were removed, cell medium changed, and cell culture continued. Degree of cell necrosis and apoptosis were measured 1 h, 1 day and 1 week following exposure and compared to control cultures that were not exposed to any agents. Fibroblast and tenocyte cultures, but not hMSC cultures, exposed to bupivacaine at 0.25% and 0.5% concentrations showed significantly greater cell necrosis than control...
