GC-MS-Bestimmung von Ethylglucuronid im Serum mit Propylglucuronid als internem Standard

Abstract

The chromatographic properties of acetyl and PFP (Perfluorpropionyl) derivatives of some synthetically prepared alkyl glucuronides on capillary columns with dimethylsilicone stationary phase and their mass spectrometric data are presented. The homologous derivatives show extensive conformity in the fragmentation patterns demonstrating that they are diacyl alkyl glucuronide lactones. The determination of ethyl glucuronide as the PFP derivative in serum after addition of 20 mg/L propyl glucuronide as internal standard is described. A parallel sample is analysed after addition of ethyl and propyl glucuronide in the same concentration of 20 mg/L so that an external calibration is not required.

Zusammenfassung

Es werden die chromatographischen Eigenschaften von Acetyl- und PFP- (Perfluorpropionyl-) derivaten einiger synthetisch dargestellter Alkylglucuronide auf einer OV-1-Kapillarsäule sowie die massenspektrometrischen Daten mitgeteilt. Die homologen Derivate zeigen weitgehende Übereinstimmung im Fragmentierungsmuster. Es wird hieraus auf die Struktur von Diacyl-alkylglucuronidlactonen geschlossen. Es wird die Bestimmung von Ethylglucuronid als PFP-Derivat im Serum unter Zusatz von 20 mg/L Propylglucuronid als innerem Standard beschrieben, wobei eine Parallelprobe nach Zusatz von 20 mg/L Propylglucuronid und Ethylglucuronid in gleicher Weise aufgearbeitet wird, so daß auf eine externe Kalibration verzichtet werden kann.
