Zusammenfassung

Die Zytogenetik spielt eine bedeutende Rolle in der genetischen Diagnostik. Auch wenn sie heute scheinbar zunehmend von modernen Techniken wie der molekularen Karyotypisierung in den Hintergrund gedrängt wird, bleibt sie besonders im Bereich der Gynäkologie und Geburtshilfe für viele Fragestellungen die entscheidende Basisdiagnostik. Chromosomenaberrationen sind die Ursache für etwa 50% aller spontanen Aborte, und bei etwa 5% aller Paare mit habituellen Aborten kann eine balancierte Chromosomentranslokation nachgewiesen werden. Zudem wird die zytogenetische Diagnostik weiterhin als einzige gesamtgenomische Untersuchungsmethode in der pränatalen Routinediagnostik eingesetzt. Für eine sinnvolle Aufklärung und Beratung der Patienten ist fundiertes Wissen über die verwendeten Techniken, ihre Möglichkeiten und Grenzen sowie die Kenntnis über die Art der zytogenetisch detektierbaren Chromosomenaberrationen und die sich daraus ergebenden klinischen Konsequenzen und Wiederholungsrisiken notwendig.

Abstract

Cytogenetic diagnostics plays a crucial role in human genetics. Although today it appears to have become substituted by modern techniques, such as molecular karyotyping, it remains the basic diagnostic approach for many questions in obstetrics and gynecology. Chromosome aberrations are the reason for approximately 50% of spontaneous abortions and balanced translocations can be found in approximately 5% of couples with recurrent abortions. Moreover, cytogenetics remains the only technique routinely applied for nondirectional whole genome analyses in prenatal diagnostics. An understanding of not only the technique, its potential, and limitations, but also a profound knowledge of the nature of detectable cytogenetic aberrations, their clinical consequences, and recurrence risks are necessary for appropriate counseling of the patients.
