Zusammenfassung
Das Melanom der Haut ist ein invasiv wachsender und frühzeitig metastasierender Tumor. Für die Ablösung einzelner Melanomzellen vom Primärtumor, Migration dieser Zellen durch Basalmembranen, Extravasation aus Blut- und Lymphgefäßen bis hin zur Bildung von Fernmetastasen ist ein komplexes Zusammenwirken von Tumorzellen, extrazellulärer Matrix (ECM) und den tumorumgebenden Bindegewebszellen notwendig. Diese Prozesse erfordern die koordinierte Expression und/oder deren Aktivierung von proteolytischen Enzymen. Neben Aspartyl- und Cysteinproteinasen sind daran insbesondere Serinproteinasen mit den Komponenten des Plasminogenaktivierungssystems (uPA, uPAR, tPA, PAI-1 und PAI-2) sowie die Matrix-Metalloproteinasen (MMP) mit ihren spezifischen Inhibitoren (TIMP) beteiligt. Die Interaktion von Melanomzellen mit Zellen oder Komponenten der ECM erfolgt über spezifische Adhäsionsrezeptoren, wie z. B. Integrine oder CD44, die z. T. auch funktionell aktive Proteinasen binden können. In dieser Übersichtsarbeit werden diese funktionellen Aspekte und ihre Bedeutung für die Melanomprogression dargestellt.
Abstract
Cutaneous melanoma is an invasive and early metastazising tumor. Melanoma cells detach from the primary tumor, penetrate the basement membrane, invade lymphatics and blood vessels, and form metastases. These processes all depend on coordinated expression and/or activation of proteolytic enzymes. In addition to aspartyl- and cysteineproteinases, serine proteinases including the plasminogen activator system (uPA, uPAR, tPA, PAI-1 and PAI-2) and matrix metalloproteinases (MMPs) with their tissue inhibitors (TIMPs) play an essential role in these processes. In addition, melanoma cells require specific adhesion molecules such as integrins and CD44 for interaction with other cells and components of the extracellular matrix (ECM); these are also involved in binding activated MMPs on the cell surface. In this review we discuss these functional aspects of melanoma progression.
