Zusammenfassung
Die regulatorische Toxikologie hat das Ziel, durch Festlegung von sicheren Standards und Normen die Bevölkerung und Umwelt vor Gefährdungen durch Chemikalien in allen Anwendungsbereichen sicher zu schützen. Verschiedene Institutionen sind mit unterschiedlichen Arbeitsschwerpunkten beteiligt: Behörden (Regulierung), Industrie (toxikologische Stoffcharakterisierung), Universität (Wirkmechanismen) und Vertragsinstitute (Aufarbeitung, Versuchsdurchführung). Der Toxikologe bearbeitet im Risikoregulierungsprozess vor allem die Schritte der Risikoabschätzung und Risikobewertung. Durch die rapiden Entwicklungen in der Molekular- und Zellbiologie werden ständig neue, innovative Testsysteme verfügbar (Invitro-Systeme, “omics”, Knock-out-Mäuse). Gleichzeitig besteht ein Zwang zur Nutzung von Ersatzmethoden. Dadurch wird die Validierung neuer Testsysteme zu einer zentralen wissenschaftlichen Herausforderung für die regulatorische Toxikologie. Darüber hinaus besteht ein kontinuierlicher Harmonierungsbedarf, vor allem auch bei Wertungsfragen (Adversität, Sicherheitsfaktoren, Niedrigdosis-Extrapolation usw.). Ein Arbeitskreis in der Deutschen Gesellschaft für Toxikologie widmet sich diesen Aufgaben.
Abstract
Regulatory toxicology aims at protecting the population and environment from toxic effects of chemicals, pesticides, food additives, cosmetics, drugs, medical products, genetic engineering products, natural toxins, etc. by establishing and applying safety standards. Different institutions are involved: authorities (regulation), industry (characterization of toxicity), universities (mechanisms of action), and contract institutes (compilation, testing). Toxicologists are involved in the regulation processes primarily at the levels of risk assessment and risk estimation. As a result of the progress in molecular and cell biology,new and innovative test methods evolve at high speed (in vitro systems, “omics,” knock out mice). At the same time there is political compulsion to employ alternative methods. Therefore, the validation of new test systems is a major scientific challenge for regulatory toxicology. Moreover, there is need for harmonization in many areas, including evaluation paradigms (adversity, safety factors, low-dose extrapolation). A study group in the German Society for Toxicology works on these tasks.
