Background and Purpose:
Due to its properties rhenium-188 (188Re) seems to be a promising radionuclide for stent coating to reduce restenosis following percutaneous transluminal angioplasty (PTA). In order to characterize the early effects of local 188Re treatment, human aortic smooth muscle cells (HaSMCs) were incubated with different doses of 188Re.
Material and Methods:
2 days after plating, HaSMCs were treated for a period of 5 days with 188Re. The total radiation doses applied were 1 Gy, 4 Gy, 8 Gy, 16 Gy, and 32 Gy. On days 1, 3, 5, and 7 (i.e., 2 days after the end of 188Re incubation), cell growth, clonogenic activity, cell migration, cell-cycle distribution, as well as matrix synthesis were evaluated.
Results:
From the 1st day on, a dose-dependent growth inhibition was observed. Cumulative doses of ≥ 8 Gy completely inhibited colony formation. The results of the migration tests were contradictory; on day 3 the migratory activity of all treated cells was increased compared to the controls, on day 5 it was reduced. Cumulative radiation doses of ≥ 8 Gy resulted in an increased fraction of cells in G2/M-phase. The synthesis of the extracellular matrix protein tenascin was not affected by the treatment.
Conclusion:
Incubating human smooth muscle cells with 188Re for a period of 5 days (i.e., seven half-lives) results in an effective inhibition of muscle cell proliferation and colony formation. Partially, this is due to a radiation-induced G2/M-phase block. Cell migration and matrix synthesis were not effectively affected in the presented in vitro system.
Hintergrund und Ziel:
Durch seine spezifischen Eigenschaften erscheint Rhenium-188 (188Re) als vielversprechendes Radionuklid für eine Stentbeschichtung, um die Restenoseentwicklung nach perkutaner transluminaler Angioplastie (PTA) zu vermindern. Zur Untersuchung der Früheffekte einer lokalen 188Re-Behandlung wurden humane aortale glatte Muskelzellen mit unterschiedlichen Dosen 188Re inkubiert.
Material und Methodik:
2 Tage nach Aussaat wurden die glatten Muskelzellen für 5 Tage mit 1 Gy, 4 Gy, 8 Gy, 16 Gy oder 32 Gy 188Re behandelt. An den Tagen 1, 3, 5 und 7 (d. h. 2 Tage nach Beendigung der 188Re-Inkubation) wurden Proliferation, klonogene Aktivität, Migration, Zellzyklus und extrazelluläre Matrixsynthese untersucht.
Ergebnisse:
Vom 1. Behandlungstag an waren dosisabhängige Wachstumsinhibitionen zu beobachten (Abbildung 2). Dosen ab 8 Gy führten zur kompletten Unterdrückung der klonogenen Aktivität (Abbildung 3). Der Einfluss auf das Migrationsverhalten war widersprüchlich; an Tag 3 migrierten die bestrahlten Zellen besser als die Kontrolle, an den anderen Tagen schlechter (Abbildung 5). Eine kumulative Dosis ab 8 Gy resultierte in einem erhöhten G2/M-Phasen-Anteil der Zellen (Abbildung 4). Die Synthese des Extrazellulärmatrixproteins Tenascin wurde durch die Behandlung nicht verändert (Abbildung 6).
Schlussfolgerung:
Eine Behandlung humaner glatter Muskelzellen mit 188Re über 5 Tage (dies entspricht sieben Halbwertszeiten mit 99,25% Dosisapplikation, Abbildung 1) resultiert in einer effektiven Inhibition der Proliferation und der Koloniebildungsfähigkeit der Zellen. Dies wird teilweise durch einen G2/M-Phasen-Arrest der bestrahlten Zellen erreicht. Die Migration und die Matrixsynthese wurden im vorgestellten In-vitro-System nicht effektiv beeinflusst.
