Zusammenfassung
In jüngster Zeit erhielten das Konzept der Probiotika und deren Einfluß auf die menschliche Gesundheit große Beachtung. Die vorliegende Arbeit untersucht die biologischen Wirkungen des probiotischen Stamms Lactobacillus acidophilus La1. In In-vitro-Studien wies La1 ausgeprägte Fähigkeiten zur Anheftung an Caco-2-Zellen auf, welche durch Kolonisierung von axenischen Mäusen mit menschlicher Mikroflora weiter bestätigt wurden. Eine Modulierung des spezifischen und unspezifischen Immunsystems wurde durch vermehrte IgA-Bildung nach oraler Impfung und Aktivierung der Phagozytoseaktivität nach Aufnahme von La1 nachgewiesen. Die Stimulierung der Phagozytose war dosisabhängig, und eindeutige Resultate wurden mit 150 ml eines fermentierten Milchprodukts (109 cfu/Tag) erzielt. Zusätzlich wurde eine kompetitive Verdrängung von potentiell pathogenen Darmkeimen, d.h. E. coli, Salmonella und Yersinia, beobachtet.
Diskussion: Zusammenfassend wurde belegt, daß La1 die natürlichen Abwehrkräfte beim Menschen verstärkt, indem die Zusammensetzung der Mikroflora sowie die spezifischen und unspezifischen Immunmechanismen verbessert werden.
Summary
Recently, the concept of probiotics and its implication for human health has emerged. This paper is focused on the biological effects of the probiotic strain Lactobacilllus acidophilus La1. In in vitro studies La1 demonstrated superior adhesion properties on Caco-2 cells, which were further confirmed by colonization of axenic mice carrying a human microflora. In human volunteers La1 showed temporary colonization upon ingestion of fermented milk products containing La1. Modulation of the specific and nonspecific immune system was demonstrated by increased IgA production following an oral vaccination and activation of the phagocylotic activity after La1 ingestion, respectively. Stimulation of the phagocytosis was dose-dependent and significant results were obtained with 150 ml of fermented milk products (109 cfu/day). In addition, competitive inhibition of potentially pathogenic enteropathogens, i.e., E. coli, Salmonella and Yersinia, was found.
Discussion: In conclusion, La1 increases the human natural defenses by improving the microflora composition and specific and nonspecific immune mechanisms.
